NF-κB Signaling and Response Durability After Ide-Cel

Single-cell RNA sequencing of 40 idecabtagene vicleucel (ide-cel) products sheds light on the role that NF-κB signaling plays in the durability of response to chimeric antigen receptor (CAR) T-cell treatment for multiple myeloma. The analyses showed that CD4 CAR-T cells generating durable responses shared a common characteristic: a transcriptional program marked by high levels of NF-κB activity, which in turn was associated with longer progression-free survival and overall survival. Identifying drivers of durable response is important for informing patient counseling and for optimizing CAR-T efficacy. There are additional implications for CAR-T manufacturing decisions, as researchers also determined that elevated NF-κB activity is a global marker of T-cell fitness that is present before CAR-T cells are even engineered.
